A Nonsecosteroidal Vitamin D Receptor Modulator Ameliorates Experimental Autoimmune Encephalomyelitis without Causing Hypercalcemia

1,25-(OH)2D3 shifts the balance from Th1 to Th2 cells in activated PBMCs. Primary cells isolated from donors were stimulated with TPA (100 ng/mL) and PHA (25 μL/mL) and treated with vehicle or various concentrations of 1,25-(OH)2D3 for 24 hours. Cytokines were measured by LINCOplex human cytokine kit on supernatants obtained from vehicle-treated or VDR ligand-treated samples using antibodies for human IL-4, IL-5, IL-10, and IFN-γ. The amount of IL-4, IL-5, IL-10, and IFN-γ protein levels is shown as mean ± SE of triplicate experiments.
