Sickle Cell Disease Activates Peripheral Blood Mononuclear Cells to Induce Cathepsins K and V Activity in Endothelial Cells

Cathepsins K and V activities induced by sickle cell disease PBMCs are significantly reduced by JNK inhibition with SP600125. HAECs were incubated with or without 10 μM of the JNK inhibitor, SP600125, 1 hour prior to TNFα stimulation, as described previously. Cocultures with AA or SS PBMCs were maintained for an additional 20 hours. Cell lysates were collected and analyzed via multiplex cathepsin zymography. Densitometric analysis quantified active cathepsins K and cathepsin V ( , , SEM bars shown).
