Schematic representation of methotrexate’s pharmacodynamic. Methotrexate (MTX) indirectly blocks the formation of dTMP by inhibiting DHF reductase. FH2, which accumulates following the inhibition of DHF reductase, in turn inhibits thymidylate synthase. DHF = dihydrofolate; FH2 = dihydrofolic acid; FH4 = tetrahydrofolic acid; dTMP = thymidine monophosphate; and dUMP = deoxyuridine monophosphate.
