Extracted nucleic acid from purified PH1 virus was treated with various nucleases and then analysed by gel electrophoresis to detect whether the nuclease digested (+) or did not digest (−) the genome. Control nucleic acids used to confirm activity of the nucleases were λ DNA, a DNA oligonucleotide, and yeast transfer RNA.
