Effects of N-Terminal and C-Terminal Polyhistidine Tag on the Stability and Function of the Thermophilic P450 CYP119

<div>Kinetic analysis of Amplex Red reaction with WT CYP119 (•), N-His-CYP119 (■), and C-His-CYP119 (△) at room temperature (a) and 65°C (b). Reactions were performed using 10 <i>μ</i>M Amplex Red, 1.5 mM H<sub>2</sub>O<sub>2</sub>, and 1.5 <i>μ</i>M enzyme in 50 mM sodium phosphate buffer at pH 7.4. The data are fit to one-phase association kinetics.</div>
