Detection of GAD65 Autoreactive T-Cells by HLA Class I Tetramers in Type 1 Diabetic Patients

Figure 1

Statistical analysis of the percentages of CD3+/CD8+/GAD65 peptide autoreactive T cells in PBMC of normal controls ( ) and T1D patients ( ). In PBMC of T1D patients, cultured with IL-2 alone for days, the median of CD3+/CD8+/GAD65 reactive T cells was 0.70 [interquartile range (0.22–1.19); range (0.00–4.3)], while when cultured with the GAD65 peptide for 4 days and IL-2 for additional 2 days, the median was 0.81 [interquartile range (0.50–4.04); range (0.14–15.6)] (Mann-Whitney U Test PBMC GAD65 peptide stimulated versus PBMC IL-2 treated, ; Wilcoxon’s Test PBMC of T1D patients versus PBMC of controls, both cultured with IL-2, ; Wilcoxon’s Test PBMC of T1D patients versus PBMC of controls, both stimulated with GAD65 peptide, ).
