Aquaporin-6 Expression in the Cochlear Sensory Epithelium Is Downregulated by Salicylates

Effect of aspirin treatment on aquaporin (AQP1, 2, 4, 6, 7) mRNA expression in mouse inner ear. Quantitative real-time RT-PCR of total RNA was performed in mouse inner ear mRNA extract under three different conditions: saline (controls), aspirin (mice treated for 3 days and sacrificed 3 hours after the last injection) recovery (treated for 3 days and sacrificed 24 hours after the last injection). Specific primers for AQP1, 2, 4, 6, 7 were used and qRT-PCR performed as indicated (see Methods). The relative mRNA levels were determined by comparative quantitation (Corbett), the values obtained were normalized to the corresponding 𝛽 -actin level and the results expressed as fold change. Results indicate that mRNA expression level of AQP6 was significantly lower in aspirin treated mice.
