Cell Density Plays a Critical Role in Ex Vivo Expansion of T Cells for Adoptive Immunotherapy

Cell density determines the fate of T cell activation. PBMCs cultured at different cell densities in 6-well plates were activated with anti-CD3 antibody alone or plus anti-CD28 antibody in the presence of IL-2. T cell activation was measured as percentage of activated T cells in culture after six days of activation by morphological criteria of T cell blasts on flow cytometer. Similar results were obtained in two independent experiments.
