Lentiviral-Mediated RNA Interference against TGF-Beta Receptor Type II in Renal Epithelial and Fibroblast Cell Populations In Vitro Demonstrates Regulated Renal Fibrogenesis That Is More Efficient than a Nonlentiviral Vector

Inhibition of α-SMA expression by TBRII RNAi in renal cell lines. Both expression vectors significantly reduced α-SMA expression in renal fibroblasts (NRK-49F) at 72 hrs indicating successful suppression of myofibroblast activation. In the epithelial cells (NRK-52E), α-SMA expression was markedly reduced with both expression constructs at both time points, indicating inhibition of EMT. The reduction in expression was most marked with the lentiviral vector. ADU: arbitrary densitometry units.
