Wild type human HMGB1 is composed of three domains: the A box (amino acid residues 1–85), the B box (amino acid residues 88–162), and the C tail (amino acid residues 186–215). A box+C tail: A box fused with C tail directly. A box+(Gly₄Ser)₃+C tail: A box fused with C tail via a (Gly₄Ser)₃ linker.
