Evaluation of the morphology and alkaline phosphatase activity of expanded reprogrammed colonies of SACK-D strains mPanstra-12 and mPanstra-25. Shown are examples of phase micrographs of colonies developed from reprogrammed cell clones and stained for alkaline phosphatase activity. The estimated alkaline-phosphatase-positive (AP+) cell fractions for colonies with epithelial (epl) or striated (str) morphology were 100% for mESCs, 10% for 25-KOSR-Xn-92 (SACK-D strain #—derivation medium—xanthine supplementation—subclone #), 50% for 12-DFBS-Xn-45, and 90% for 12-KOSR-Xn-89. μm.
