HDAC Activity Is Required for Efficient Core Promoter Function at the Mouse Mammary Tumor Virus Promoter

Transcription dependent on core promoters is repressed by TSA. Luciferase reporter constructs were generated which contain core promoters (TATA to Inr sequences) from several viral and cellular genes, including Cytomegalovirus (CMV), c-fos, interleukin 4 (IL4), and carbonic anhydrase II (CA2). These constructs were transiently transfected into either Hela or 1470.2 cells which were treated with or without TSA (50 ng/ml) for 6 h. Cell lysates were assayed for luciferase activity, and the data was processed as described in the legend to Figure 1. Results from at least 3 independent experiments done with Hela or 1470.2 cells are shown. Error bars represent SEM.
