Validation of the 2D-DIGE results by western blot analysis. (a) Triplicate synaptosomal preparations from wild-type and FDDKI mouse synaptosomes were separated on SDS-PAGE gels and transferred to membranes for western blot analysis with antibodies against the indicated proteins. Actual downregulation in the synaptosomal samples of FDDKI mice can be observed for Dlg4/Psd95 and Dusp3 proteins. α-Tubulin antibody was used to show equal loading of the protein extracts. (b) Total brain homogenates from wild-type and FDDKI mice were analyzed by western blot with antibodies against Dusp3. Actin antibody was used to show equal loading of the protein extracts.
