Gene Therapy of Multiple Sclerosis Using Interferon β-Secreting Human Bone Marrow Mesenchymal Stem Cells
Figure 4
MSCs-IFNβ modulate immune responses in EAE mice. (a) The serum was isolated from the three groups of EAE mice 30 days after PBS-, MSCs-GFP, or MSCs-IFNβ treatment (/each group). Cytokine level of Th1 (IFN-γ and TNF-α) and Th2 (IL-4 and IL-10) in the serum was quantified by ELISA. (b) Splenocytes (5 × 105/well) isolated from EAE mice at day 37 after immunization (/each group), and then the cells were stimulated with MOG35–55. The number of MOG-specific IFN-γ /IL-4 producing splenocytes was determined by ELISPOT assay. Representative well show spots for IFN-γ /IL-4 production. Magnification: ×1. Columns: mean; bars: SE. *; **; Student’s t-test. The results are representative of three independent experiments.
