Silencing of Entamoeba histolytica Glucosamine 6-Phosphate Isomerase by RNA Interference Inhibits the Formation of Cyst-Like Structures

Expression of chitin and viability of Gln6Pi silenced trophozoites after CLS induction and detergent treatment. Untransfected trophozoites or transfected with 40  g of si 154-Gln6Pi or 40  g of each si 154-Gln6Pi and si 229-Gln6Pi (siRNA mix 40 ug) were induced to CLS followed by treatment with 0.5% sarkosyl. Cells were stained with FDA for viability before ((a), (d), and (g)) and after ((c), (f), and (i)) CLS induction and detergent treatment. Calcofluor white staining is shown for cells after CLS induction and detergent treatment ((b), (e), and (h)). Viable cells are observed in green and chitin-positive cells are whitish blue under UV light microscopy.
