Effect of Cyclic Mechanical Stimulation on the Expression of Osteogenesis Genes in Human Intraoral Mesenchymal Stromal and Progenitor Cells
Figure 4
Influence of mechanical stimulation on alkaline phosphatase (ALP) expression and demonstration of mineralized matrix using ARS staining of human intraoral MSPCs. (a) Representative pictures of unstained (EX(−) and EX(+)) and osteogenic differentiated (OG(−) and OG(+)) MSPCs. Quantitation of calcium deposited into the cultures revealed a significant increase in the 570 nm OD in osteogenic differentiated and mechanically stimulated MSPCs after (b) 7 days and (c) 14 days, when compared to undifferentiated controls. (d) Quantitation of ALP enzymes revealed a significant increase compared to corresponding controls. EX(−) represents the undifferentiated mechanically unstimulated group, EX(+) the undifferentiated mechanically stimulated group, OG(−) the osteogenic differentiated mechanically unstimulated group, and OG(+) the osteogenic differentiated mechanically stimulated group. The values are of independent experiments performed in triplicate (); *.
