Calibration curves of H₂O₂ in the assay system. H₂O₂ concentration was used in the range of 1 to 1000 μM (a) and 1–15 μM (b) with 1 h incubation of the reaction mixture. Time kinetics (0–2 h) for completion of reaction for two concentrations of H₂O₂ (100–200 μM) without (c) and with (d) TCA treatment of H₂O₂ followed by neutralization prior to use. Dose curve for H₂O₂ (1–16 μM) in both TCA treated and nontreated condition with 2 h incubation of the reaction mixture (e). In all cases 125 μM HVA and 0.1 U/mL HRP were used in 2 mL reaction mixture at 25°C.
