The sample (middle) is Kohler illuminated using a high numerical aperture phase mask, resulting in annular illumination at a high angle of incidence. The objective lens, right, is not a phase objective but has a numerical aperture large enough to capture the unscattered annular illumination. We used a phase mask designed for a 40x/0.55 phase contrast objective and a 10x/0.25 objective lens to collect images.
