Hyaluronan-Phosphatidylethanolamine Polymers Form Pericellular Coats on Keratinocytes and Promote Basal Keratinocyte Proliferation

HA-PE simulates pericellular coat formation in cultured fibroblasts. 500 kDa HA was linked to PE using EDC to achieve a stoichiometry of 1 : 10 (HA : PE). (a) This ratio was selected because it resulted in the largest pericellular HA coats, which were detected using fluorescent particle exclusion assays. (b) The black patches on the epifluorescent images are areas of pericellular coats. A Hofmann image is included which shows cells at the center of the pericellular coats. Images were taken with a 10X objective. The percentage cells/10x field that were surrounded by HA coats were calculated using Hofmann optics. (c) Values are Mean and S.E.M fields.
