Oxidative Stress Induces Endothelial Cell Senescence via Downregulation of Sirt6

Sirt6 overexpression inhibits the activation of senescence pathways induced by H₂O₂. ECs were infected with adenovirus carrying Sirt6 gene (Ad-Sirt6) or empty vector (control, Ad-Con). At 1 day after transduction, cells were exposed to 75 𝜇M H₂O₂ or vehicle (H₂O). At 1 day after H₂O₂ treatment, cells were lysed and phosphorylated-Rb, p53, and p21 were determined by Western blot.
