Effect of serum deprivation and induction as well as hypoxia on Nampt/PBEF/visfatin expression in colon adenocarcinoma cell line SW620. (a) Effect of serum withdrawal and serum induction. In a serum withdrawal experiment, cells were grown with 10% FBS to 80% confluence, at which point serum supplementation was withdrawn for 12, 24, and 48 hours; data are presented as relative expression of Nampt (mean ± SD of three independent biological experiments) in starved versus FBS-supplemented cells. In a serum induction experiment, cells were grown with 10% FBS to 80% confluence at which point serum supplementation was withdrawn and resupplemented after 24 hours for 24 or 48 hours in all cultures except for controls; data are presented as relative expression of Nampt (mean ± SD of independent biological experiments) in resupplemented versus FBS-starved cells. (b) Effect of hypoxia. Cells were grown for 80% confluence and stimulated with 50, 100, and 200 μM concentrations of cobalt chloride, a hypoxia-mimetic agent, for 8, 24, and 48 hours. Data are presented as relative expression of Nampt (mean ± SD of three independent biological experiments) in stimulated compared to unstimulated cells.
