ERK1/2 mediated SAA-induced Visfatin production. (a) RAW264.7 cells were stimulated with 50 μg/mL SAA for different times (0 min, 5 min, 15 min, 30 min, 1 h, and 2 h). The phosphorylation of ERK1/2 was detected with western blot. (b) RAW264.7 cells were preincubated with PD98059 (20 μmol/L) and then treated with 50 μg/mL SAA for 24 hours. The expression of Visfatin was detected with western blot at protein level. (c) After pretreatment with FPR2 siRNA (20 nM, for 6 h) and WRW⁴ (30 μM, 1 h) before SAA (50 μg/mL) for another 24 h, or stimulation of WKYMVm (100 nM) for 24 h, phosphorylation of ERK1/2 was measured with western blot. versus control group; versus SAA treatment group; versus SAA treatment group.
