Inhibitory Effects of Ethyl Acetate Extract of Andrographis paniculata on NF-κB Trans-Activation Activity and LPS-Induced Acute Inflammation in Mice

Effect of the EtOAc extract of AP on NF-κB dependent luciferase reporter activity in Raw264.7 cells activated by LPS/IFN-γ (L/I). Raw264.7 macrophages transient transfected with a NF-κB reporter plasmid were pretreated with 2.5–20 μg ml⁻¹ of AP EtOAc extract or helenalin (NF-κB inhibitor, 10 μM) for 1 h and then stimulated with LPS 100 ng ml⁻¹/IFN-γ 1000 U ml⁻¹ for 8 h. The activity of NF-κB was estimated by the Dual-Glo Luciferase reporter assay. The data are expressed as the mean ± SD from three independent experiments.
