Leea indica Ethyl Acetate Fraction Induces Growth-Inhibitory Effect in Various Cancer Cell Lines and Apoptosis in Ca Ski Human Cervical Epidermoid Carcinoma Cells

Figure 9

Inhibitory effect of Ac-DEVD-CHO on LIEAF-induced apoptosis in Ca Ski cells. Ca Ski cells were pretreated without or with 50 μM Ac-DEVD-CHO for 1 hour prior to the treatment of 500 μg/mL LIEAF for 24 hours, and then apoptosis was performed by TUNEL and PI staining. For annexin V-PI staining, cells were cultured in the absence or presence of 50 μM Ac-DEVD-CHO for 1 hour before being stimulated with 500 μg/mL LIEAF for 72 hours. (a) Flow cytometric analysis of TUNEL assay. (b) Histograms showing the appearance of hypodiploid sub-G1 cells (arrow) detected by PI staining. (c) Flow cytometric fluorescence patterns of annexin V-PI staining. (d) Summarized results for TUNEL, PI, and annexin V-PI staining. Data showed means ± S.E. of three experiments. The asterisk represented significantly different value from respective group (* ).
