Inhibitory effect of DEN3 on the phosphorylation of Akt. B16F10 cells were cultured in various concentrations of DNE3 for 24 hours, and then cell lysates were subjected to SDS-PAGE followed by Western blotting with anti-PI3K (a), anti-phospho-Akt (b), anti-phospho-p38 (c), and anti-phospho-ERK1/2 (d) antibodies. Signals of proteins were visualized with an ECL detection system. Determined activities of these proteins were subsequently quantified by densitometric analysis with that of control being 100% (defined as 1.0) as shown just below the gel data. Results were statistically evaluated by using one-way ANOVA with post hoc Dunnett's test (***).
