PCE upregulated ROS level in Bel-7402 cells. (a) After 24 h PCE (100–400 μg/mL) treatment, intracellular ROS production in suspension-cultured Bel-7402 cells was detected as described in Materials and Methods section. ROS level was expressed as fold activation over control. For ROS inhibition, suspension-cultured Bel-7402 cells were pretreated with NAC (50 mmol/L for 2 h), followed by PCE (100–400 μg/mL) treatment for 24 h, and subjected to apoptosis (b), caspase-3 (c), and caspase-9 (d) activity assay. Caspases activities were expressed as fold activation over control.
