GJH downregulates the expression of ICAM-1 through reduction of the expression and transcriptional activity of NF-κB. (a) GJH significantly reduces the expression of ICAM-1 in both dose- and time-dependent manners, as shown by western blot analysis. (b) Western blotting reveals that GJH reduces the amounts and the nuclear translocation of NF-κB p65 and p50 subunits in the cells. (c) GJH reduces the DNA-binding activity of NF-κB to ICAM-1 as shown by EMSA analysis. (d) GJH downregulates TNF-α-dependent and TNF-α-independent ICAM-1 expression in part through NF-κB as demonstrated by the luciferase reporter assay.
