Role of PI3K and MAPK signalling in anandamide-mediated protection against 6-OHDA toxicity. (a) PC12 cells were pre-treated with 40 M LY294002 for 1 hours prior to addition of 25 M anandamide for 24 hours followed by treatment with 100 M 6-OHDA for a further 24 hours. Values represent the mean ± SEM of 4 independent experiments. (b) PC12 cells were treated with 25 M anandamide for 0–24 hours. The phosphorylation state of ERK1/2 was analysed by Western blotting. Actin protein levels were also analysed as a loading control. The results are representative of two separate experiments. (c) 10 M U0126, the MAPK pathway inhibitor was added 1 hour prior to incubation of PC12 cells with 25 M anandamide for 24 hours and with 100 M 6-OHDA for a further 24 hours.
