Cysteine-10 on 17<i>β</i>-Hydroxysteroid Dehydrogenase 1 Has Stabilizing Interactions in the Cofactor Binding Region and Renders Sensitivity to Sulfhydryl Modifying Chemicals

<table>Preincubation with NADPH protects 17<i>β</i>-HSD1 from inhibition by NEM. Lysates of HEK-293 cells expressing 17<i>β</i>-HSD1 were preincubated for 15 min with saline or various concentrations of NADPH as indicated, prior to the addition of estrone (200 nM), NADPH (400 <i>μ</i>M), and the sulfhydryl modifying agent N-ethylmaleimide (NEM, 60 <i>μ</i>M). The estradiol produced was determined after 10 min of incubation. Data (mean ± SD) were normalized to the control in the absence of NEM and were obtained from at least three independent experiments measured in triplicate.</table>
