Comparison of the Pharmacological Effects of Paricalcitol and Doxercalciferol on the Factors Involved in Mineral Homeostasis

Effects of paricalcitol and doxercalciferol on serum PTH and PTH mRNA levels in uremic rats. The 5/6 NX rats on a hyperphosphatemia-inducing diet were treated with vehicle, paricalcitol, or doxercalciferol at 0.33  g/kg, i.p., 3 times/week, for two weeks ( = 8–11 per group). Blood samples and parathyroid gland were collected for the measurement of serum PTH (a) and PTH mRNA (b). For (b), real-time RT-PCR was performed as described in Materials and Methods. The mRNA expression level was first normalized to the GAPDH mRNA level and then calculated as % of control (Sham-vehicle, 100%). One way ANOVA Dunnett test with 95% confidence intervals of difference was performed for statistical comparisons. , versus Sham. , versus 5/6 NX-Vehicle.
