Wogonin Attenuates Ovalbumin Antigen-Induced Neutrophilic Airway Inflammation by Inhibiting Th17 Differentiation

Suppression of IL-17 by wogonin treatment in the human MLR. For the human two-way MLR (a), wogonin (1 μM) was exposed to a mixture of human allogeneic lymphocytes and then incubated for seven days. Seven days after incubation, the supernatants were analyzed using ELISA to detect the typical cytokines of IFN-γ (center), IL-4 (right), and IL-17 (left) for Th1, Th2, and Th17, respectively. For the human one-way MLR (b), wogonin (1 μM) was incubated with human Mo-DCs for two days. Then, the wogonin-treated human Mo-DCs were incubated with human allogeneic naïve CD4+ T cells in the absence of wogonin for seven days. The cells were restimulated with anti-human CD3 and CD28 antibodies for 24 h, and the supernatants were collected for ELISA to detect the typical cytokines of IFN-γ (center), IL-4 (right), and IL-17 (left) for Th1, Th2, and Th17, respectively.
