Diagram of extant small RNA cloning strategies. Following small RNA enrichment, all strategies share the same outline of first placing an adaptor on the end of the target RNAs, then placing a second adaptor on the end of the RNAs, followed by reverse transcription, amplification and cloning. Recent advances in next generation high throughput single molecule sequencing platforms have eliminated the actual cloning step but still relie to a greater or lesser extent on the same upstream methods.
