Suppression of HSP70 sensitizes TGF- signaling. (a) NCI-H292 cells were transfected with HSP70 siRNA or control siRNA and SBE4-luc, and then treated with TGF- () for . Luciferase activity was measured by a luminometer. (b) Cells transfected with HSP70 siRNA or control siRNA treated with or without TGF- () for and stained with the PI DNA dye were subjected to cell cycle analysis using FACS. The data were expressed as a histogram with the DNA content (FL2) along the -axis. (c) Cells were transfected with HSP70 siRNA or control siRNA treated with or without TGF- () for indicated time. Cell lysates were immunoblotted with antibody specific to c-Myc, phospho-Smad2, or HSP70.
