Myosin VI and Associated Proteins Are Expressed in Human Macrophages but Do Not Play a Role in Foam Cell Formation in THP-1 Cells

Figure 10

Effect of siRNA targeted to Myo6 on the uptake of LDL and oxLDL by THP-1 macrophages. (a), (b) THP-1 macrophages were incubated with nLDL or oxLDL (50 μg protein/mL) for 6 h (a) or time points up to 24 h (b), and the area of fluorescent signal associated with the cells was assessed using a Leica inverted DMIRB wide-field fluorescence microscope. (c), (d) THP-1 cells were transfected with siRNA targeting Myo6 or with a nonsilencing scrambled siRNA sequence (control) using HiPerFect transfection reagent. After 48 h, cells were treated with 50 μg protein/mL DiI-labelled nLDL (c) or oxLDL (d) and fixed at time points up to 24 h. Wide-field images were taken using a Leica DMRIB fluorescence microscope. The mean intensities of the fluorescent signals ((b)–(d)) were measured using Volocity software. Data shown are the means from three separate experiments, and error bars show the SEM. Significance limits, * versus nLDL (two way ANOVA).
