Relative distributions of AP-2 and Myo6 or Dab2 in THP-1 macrophages. THP-1 macrophages were fixed and probed using double immunofluorescence for (a) AP-2α2 (secondary antibody conjugated to FITC (green)) and Myo6 (secondary antibody conjugated to Alexa555 (red)); (b) AP-2α2 (secondary antibody conjugated to FITC (green)) and Dab2 (secondary antibody conjugated to Alexa555 (red)). Z-stacked images were taken on a Leica SP5 confocal microscope. Maximum projections are shown on the left for orientation. Series labeled (i) show single confocal sections and vertical xz and yz sections through the centre of the cells indicated to the left. The individual channels for Myo-6 or Dab2 (red), AP-2α2 (green), and the overlay are shown side-by-side. Enlargements of the single confocal slice, showing the plasma membrane, are shown in series (ii) and show the regions indicated in (i).
