Mutant MGMT-mediated drug selection. MGMT encodes O6-alkylguanine-DNA alkyltransferase (AGT). (a) AGT repairs O6-alkylguanine lesions in DNA resulting from exposure to methylating or chloroethylating agents, such as, temozolomide and 1,3-Bis(2-Chloroethyl)-Nitrosourea (BCNU), respectively. (b) O6-benzylguanine (BG) irreversibly inactivates wild-type AGT, sensitizing cells to O6-alkylating agents. (c) Specific AGT point mutations, including P140K and G156A, are resistant to BG inactivation but maintain normal repair activity for O6-alkylguanine lesions.
