Effects of stathmin overexpression ((a) and (b)) and knockdown ((c) and (d)) on hypoxia-induced HIF-1α and VEGF expression in OVCAR-3 cells.(a) Cells were transfected with an empty (Mock) or stathmin expression (STMN) vector and then cultured under hypoxic conditions for 5 h. (b) Cells were treated with control (–) or stathmin (+) siRNA and then cultured under normoxic or hypoxic conditions for 5 h. ((a) and (c)) Cell lysates were subjected to immunoblot analysis for HIF-1α and stathmin. The same blot was probed, stripped, and then reprobed with each antibody. The level of β-actin was used as a loading control. ((b) and (d)) Total RNA was extracted and subjected to semiquantitative RT-PCR to analyze stathmin, VEGF, and HIF-1α mRNA expression.
