Local Gene Delivery System by Bubble Liposomes and Ultrasound Exposure into Joint Synovium

Luciferase expression in HFLS transfected with bubble liposomes and ultrasound exposure compared with Lipofectamine 2000. pDNA (pCMV-Luc) and BL were mixed together with culture medium and added to the HFLS. The cells were immediately exposed to US (frequency, 2 MHz; duty, 50%; intensity, 2.5 W/cm2; US exposure time, 10 sec.). The cells were washed and cultured for 2 days, and then luciferase activity was determined as described in Section 2. The transfection of pDNA by LF2000 was also performed according to the manufacturers’ instructions. All data are shown as the (). versus other treatment groups.
