Osmolarity and Glucose Differentially Regulate Aldose Reductase Activity in Cultured Mouse Podocytes

Time course of the changes in podocytic aldose reductase (AR) activity. Podocytes were cultured for indicated time periods in media with normal (5.6 mM, NG) or high (30 mM, HG) glucose content, and/or normal (285, Nosm) or high (385, Hosm) osmolarity. Cell lysates were prepared as indicated in Materials and Methods. Enzyme activities were calculated from the rate of the decrease in NADPH absorbance and are shown as absolute values. Data are expressed as means ± SE from 3–5 independent experiments, * versus NG-Nosm cells.
