Inhibition of IAPP Aggregation and Toxicity by Natural Products and Derivatives

Dye leakage induced upon hIAPP addition (2 μM) to 0.2 mg/mL POPC/POPG (7 : 3 molar ratio) lipid vesicles in the presence and absence of indicated flavanols. Membrane disruption is detected by concentration dependent quenching of a vesicle-encapsulated dye. Within the vesicle, the carboxyfluorescein dye is at a high concentration and is quenched. Disruption of the membrane integrity of the vesicle dilutes the concentration of the dye and thereby increases the measured fluorescence.
