Gene expression analysis and viability of huPBLs. mRNA expression of (a) TLRs in huPBLs following 24 and 72 hours stimulation with CT. Quantitative real-time PCR was performed using predesigned TaqMan gene expression assays. Results show data of three separate experiments each performed in duplicates. (b) Viability of huPBLs as determined by flow cytometric PI staining. Prior to analysis, huPBLs were stimulated with increasing doses of CT and cultured for 24 hours, 72 hours, and 7 days.
