Alpha/Beta T-Cell Depleted Grafts as an Immunological Booster to Treat Graft Failure after Hematopoietic Stem Cell Transplantation with HLA-Matched Related and Unrelated Donors

Figure 1

Characteristics of -depleted graft. Apheresis products were analyzed by flow cytometry before and after T-cell depletion. (a) A representative plot of T-cell frequency of the graft before and after depletion gated from CD45+CD3+ cells. (b) Analysis by flow cytometry was performed on four out of five -depleted graft samples (the sample from the fifth patient had too few cells for analysis). Subgroups (Vδ1, Vδ2, Vγ9, and V24)+ cells of CD3+ T-cells were analyzed and representative plots from the subsets are shown. Vδ1, Vδ2, and Vγ9 subsets are shown from patient 1567 and V24 subset from patient 1620. (c) Representative plots of B- and NK-cells in the graft after depletion, gated from CD45+CD19+ or CD45+CD16+/CD56+, respectively. (d) Log depletion of T-cells and yield/recovery of T-cells after depletion.
