Polyclonal Recipient nTregs Are Superior to Donor or Third-Party Tregs in the Induction of Transplantation Tolerance

nTregs are superior to iTregs in the induction of mixed chimerism. Groups of B6 mice were grafted with 15–20 × 106 Balb/c BM cells under the cover of costimulation blockade (anti-CD154, CTLA4Ig) and rapamycin and were additionally treated with decreasing numbers of recipient-derived nTregs (white bars) and recipient-derived iTregs (grey bars), respectively. Percentages of successfully induced chimeras are shown. Mice were considered chimeric if donor cells were detectable by flow cytometry within both the myeloid lineage and at least one lymphoid lineage for the length of follow-up. Data are pooled from multiple independent experiments (n.d.: not done).
