Targeted integration at the endogenous XPC4 locus: experimental design. The XPC4 target sequence is located in exon 9 of the XPC gene. Exons 8, 9, and 10 are shown in white boxes. Cleavage of the native XPC gene by the meganuclease yields a substrate for homologous recombination, which may use the repair plasmid depicted.
