Alkyne modification does not interfere with digestion and ligation reactions. Lanes 4 and 5 show the successful self-ligation of 371 bp alkyne DNA after an EagI digest and a PspOMI digest, respectively, which each generate 738 bp fragments. Self-ligations of EagI- and PspOMI-digested control DNA (no alkyne modification) are shown in lanes 2 and 3, respectively. Lane 1 contains a 1 kb ladder, with numbers on the left indicating selected fragment lengths in base pairs. Alkyne DNA runs slightly slower than unmodified DNA.
