The Human Stratum Corneum Prevents Small Gold Nanoparticle Penetration and Their Potential Toxic Metabolic Consequences

NAD(P)H effects from MPT-FLIM imaging. To assess the metabolic impact of AuNP penetration, NAD(P)H signals were analysed using 𝛼 1 % / 𝛼 2 % ratio and total NAD(P)H lifetime intensity using MPT-FLIM. The 𝛼 1 % / 𝛼 2 % ratio (a) is inversely related to the metabolic rate and was calculated for each of the treatment groups at 24 hours from stratum granulosum, spinosum, and basale. The total NAD(P)H lifetime intensities of selected area (b) were examined as a function of depth in non-furrow-containing regions. The beginning of viable epidermis is at 0 μm.
