Ptch recombination and expression in and mice. (a) The efficiency of loxP recombination at the Ptch locus in DNA derived from tail skin from untreated and tamoxifen treated mice was determined by allele-specific real-time PCR as described in [85]. (b) Ptch transcripts in different tissues were analyzed by RT-PCR. The transcripts derived from the and the locus (the latter equates to the floxed Ptch locus after Cre-mediated excision) were analyzed by semiquantitative RT-PCR as described in [85]. In the skin, untreated mice only expressed transcripts and the normally occurring transcript lacking exon 10. In contrast, untreated mice expressed transcripts and transcripts lacking exon 10 (asterisk) in the skin.
