Effect of intracellular Ang II on Stat3 phosphorylation. Mesangial cells incubated with 5 mM glucose (NG) containing 1 μM exogenous Ang II (NG + ex-Ang II) for 20 minutes showed a significant increase in both tyrosine 705 and serine 727 Stat3 phosphorylation compared to NG control. In contrast, transfection of cells with 1 μM Ang II (NG + t-Ang II) for 20 minutes increased Stat3 phosphorylation only at tyrosine 705 but not at serine 727 residue.
