Caspase-10 Is the Key Initiator Caspase Involved in Tributyltin-Mediated Apoptosis in Human Immune Cells

Caspase-10 inhibition prevents TBT-induced apoptosis in all variants of Jurkat A3 cells. Jurkat cells were pretreated with the caspase-10 inhibitor AEVD (8.7 μM, 1 h) before TBT was added (1 μM, 4 h). Then, the cells were stained with annexin V-FITC/PI to separate apoptotic cells from necrotic and viable cells by flow cytometry. The histograms show the fluorescence intensity of PI-negative only (compare lower two quadrants in Figures 1(g) and 1(h)). Numbers given for TBT-treated samples represent the percentage of total cells counted (10 000). Presented are the histograms of all three Jurkat A3 variants.
