PKCδ Regulates Translation Initiation through PKR and eIF2α in Response to Retinoic Acid in Acute Myeloid Leukemia Cells

ATRA and ATO induce p-eIF2α during monocytic differentiation of AML cells. (a) ATRA (1 μM) induces monocytic differentiation in U937 and THP-1 cells but not in NB4 cells as indicated by induction of CD14 expression on the cells detected by FACS analysis. (b) ATRA and ATO induce phosphorylation (Ser51) of eIF2α expression in U937 AML cells. The U937 cells were treated with either ATRA (1 μM) or ATO (0.4 μM) for the indicated time periods and analyzed by Western blot. (c) ATRA (1 μM) also induces phosphorylation (Ser51) of eIF2α expression in THP-1 monocytic AML.
